Proteinase K is a highly active serine protease. Its enzyme activity is stable in the presence of calcium salts, and increases when a protein denaturant such as SDS or urea is also present. Proteinase K is compatible with a wide range of substrates. It preferentially degrades estel and peptide bands located next to a C-terminal hydrophobic, sulfuric, or aromatic amino acid.

2. Preparation of chromosomal DNA for in situ hybridization, fingerprinting, colony hybridization, plaque hybridization and pulsed-field electrophoresis.
